目的检测细胞外基质金属蛋白酶诱导因子(extracellular matrix metalloproteinase inducer,EMMPRIN)、基质金属蛋白酶-9(matrix metalloproteinases-9,MMP-9)在胃癌中的表达,观察其与胃癌临床病理因素的关系及两者表达的相关性,探讨EMMPRIN、MMP-9在胃癌发生、发展过程中的作用。方法应用免疫组化(Elivision plus)法检测120例胃癌及癌旁正常组织中EMMPRIN及MMP-9蛋白的表达。结果癌旁组织EMMPRIN、MMP-9蛋白表达的阳性率21.7%(26/120)和19.2%(23/120)明显低于相应胃癌组织79.2%(95/120)和82.5%(99/120),两者比较差异性显著(P<0.01),其表达水平与患者的性别、年龄无关(P>0.05),TNM分期Ⅰ、Ⅱ期胃癌组织EMMPRIN、MMP-9蛋白表达水平低于Ⅲ、Ⅳ期胃癌组织(P<0.05);高分化、中分化癌组织EMMPRIN、MMP-9蛋白的表达低于低、未分化癌组织(P<0.05);无淋巴结转移的胃癌组织EMMPRIN、MMP-9蛋白表达低于有淋巴结转移的胃癌组织(P<0.05),胃癌浸润程度越深表达越高(P<0.05);②EMMPRIN蛋白表达强度与MMP-9蛋白表达强度之间呈明显正相关(r=0.591,P<0.001)。结论 EMMPRIN、MMP-9的表达与胃癌生物学行为密切相关,两者在胃癌浸润转移过程中可能存在协同效应。EMMPRIN、MMP-9可作为胃癌侵袭转移的判断指标。
Objective To detect the expression of extracellular matrix metalloproteinase inducer (EMMPRIN) and matrix metalloproteinases-9 (MMP-9) in gastric carcinoma and to observe its relationship with clinicopathological factors The expression of EMMPRIN, MMP-9 in the occurrence and development of gastric cancer. Methods The expressions of EMMPRIN and MMP-9 in 120 cases of gastric cancer and adjacent normal tissues were detected by Elivision plus method. Results The positive rates of EMMPRIN and MMP-9 in cancer tissues were significantly lower than those in corresponding gastric cancer tissues (21.7% (26/120) and 19.2% (23/120)), which were significantly lower than those in corresponding gastric cancer tissues (79.2%, 95/120 and 82.5%, 99/120) (P <0.01). The expression level of EMMPRIN and MMP-9 in TNM stage Ⅰ and stage Ⅱ gastric cancer tissues was lower than that in stage Ⅲ and Ⅳ (P <0.05). The expression of EMMPRIN and MMP-9 in well-differentiated and moderately differentiated cancer tissues was lower than that in low-grade, undifferentiated carcinoma tissues (P <0.05). The expressions of EMMPRIN and MMP-9 in gastric cancer tissues without lymph node metastasis (P <0.05). (2) The expression of MMPMPRIN protein was positively correlated with the expression of MMP-9 protein (r = 0.591 , P <0.001). Conclusion The expressions of EMMPRIN and MMP-9 are closely related to the biological behavior of gastric cancer. There may be synergistic effects between the two in the invasion and metastasis of gastric cancer. EMMPRIN, MMP-9 can be used as indicators of invasion and metastasis of gastric cancer.
