目的研究果蝇zeste基因增强子同源物2(EZH2)和结缔组织生长因子(CTGF)基因在肺腺癌中的表达和意义。方法采用逆转录-聚合酶链反应(RT-PCR)和免疫组织化学染色法分别检测60例肺腺癌标本及相应的癌旁正常肺组织中EZH2和CTGF基因的mRNA和蛋白表达,分析EZH2和CTGF表达与肺腺癌临床参数之间的关系。结果与CTGF的表达相反,EZH2基因的mRNA和蛋白水平在肺腺癌癌组织中的表达明显高于癌旁正常肺组织(P<0.01),在低分化肺腺癌标本中的表达高于高分化腺癌(P<0.05),在晚期肺癌中的表达高于早期肺癌(P<0.05)。EZH2和CTGF的表达均与患者的性别、年龄无关(P>0.05),与吸烟有关(P<0.05)。结论 EZH2和CTGF可能分别作为癌基因和抑癌基因参与肺腺癌的发生、发展。
Objective To study the expression and significance of zeste gene enhancer homolog 2 (EZH2) and connective tissue growth factor (CTGF) genes in lung adenocarcinoma. Methods The mRNA and protein expressions of EZH2 and CTGF in 60 specimens of lung adenocarcinoma and corresponding normal lung tissues were detected by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemical staining respectively. Results In contrast to the expression of CTGF, the mRNA and protein levels of EZH2 in lung adenocarcinoma tissues were significantly higher than those in adjacent normal lung tissues (P <0.01), and were higher in poorly differentiated lung adenocarcinoma Differentiated adenocarcinoma (P <0.05), the expression in advanced lung cancer was higher than that in early stage lung cancer (P <0.05). The expression of EZH2 and CTGF were not related to sex and age (P> 0.05), but related to smoking (P <0.05). Conclusion EZH2 and CTGF may participate in the occurrence and development of lung adenocarcinoma respectively as oncogenes and tumor suppressor genes.
