目的研究橄榄油对新生期双酚A(bisphenol A,BPA)暴露后青春期雄性小鼠生殖毒性的拮抗作用。方法将32只清洁级新生ICR雄性小鼠随机分为4组,分别为对照(玉米油)组、橄榄油组、BPA(100μg/kg,玉米油稀释,前期实验中能明显引起精母细胞减数分裂阻滞)组、橄榄油+BPA(100μg/kg)组,每组8只。自出生第一天(PND 1)至PND 21(断乳),仔鼠每天进行颈背部皮下注射染毒,染毒容量为4μl/g,每天1次,连续染毒至断乳(PND 21)。PND 30,测定睾丸曲细精管直径、生精上皮厚度、管腔直径以及睾丸组织中Boule mRNA和蛋白的表达水平。结果与对照组相比,BPA组小鼠睾丸曲细精管直径和生精上皮厚度均显著下降,曲细精管管腔直径显著上升,差异均有统计学意义(P<0.05);虽然橄榄油+BPA组与对照组相比仔鼠曲细精管直径和生精上皮厚度也有明显下降(P<0.05),但与BPA组相比,曲细精管直径和生精上皮厚度均显著上升(P<0.05),曲细精管管腔直径显著下降(P<0.05)。与对照组相比,BPA组小鼠睾丸组织中Boule蛋白的表达下降,差异均有统计学意义(P<0.05)。橄榄油+BPA组小鼠睾丸组织中Boule蛋白的表达水平高于BPA组,差异有统计学意义(P<0.05)。与对照组相比,BPA组和橄榄油+BPA组小鼠睾丸组织中Boule mRNA的表达水平均降低,差异有统计学意义(P<0.05)。但橄榄油+BPA组小鼠睾丸组织中Boule mRNA的表达水平高于BPA组,差异有统计学意义(P<0.05)。结论橄榄油对BPA所致雄性小鼠生殖毒性及Boule表达下降有一定的拮抗作用。
Objective To study the antagonistic effect of olive oil on reproductive toxicity of adolescent male mice exposed to bisphenol A (BPA) in neonatal period. Compared with the control group, the expression of Boule protein in BPA group decreased significantly (P <0.05). The level of Boule protein in testis tissue of olive oil + BPA group was higher than that of BPA group (P <0.05). Compared with the control group, the expression levels of Boule mRNA in the testis tissue of BPA group and olive oil + BPA group were significantly decreased (P <0.05). However, the expression of Boule mRNA in testis tissue of olive oil + BPA group was higher than that of BPA group (P <0.05). Conclusion Olive oil can antagonize the reproductive toxicity and the decrease of Boule expression in male mice induced by BPA.
