目的探讨利用酶切保护PCR分析(EPM-PCR)检测鱼肉中二吨恶英类化合物(DLCs)的毒性当量(TEQs)的可行性。方法将2份1g鱼肉样本经过硝化、提取及净化步骤以后,采用EPM-PCR分析方法检测其中TEQs,并与高分辨率气相色谱-高分辩率质谱联用技术(HRGC-HRMS)的检测结果进行比较。结果两份鱼肉样本经酶切保护PCR分析得到的TEQs值分别是10.92pg/g和39.61pg/g,相当于HRGC-HRMS标准方法的2.91倍和3.68倍。结论EPM-PCR可作为有效的筛选鱼肉样本中DLCs的一种手段。
Objective To investigate the feasibility of using enzyme digestion protection PCR (EPM-PCR) to detect the toxic equivalents (TEQs) of two tons of dioxin in fishmeal. Methods Two samples of 1g fish were subjected to nitrification, extraction and purification steps. The results of the detection of TEQs by EPM-PCR were compared with those of high-resolution gas chromatography-high resolution mass spectrometry (HRGC-HRMS) Compare Results The TEQs of the two fish samples were 10.92 pg / g and 39.61 pg / g, respectively, which were 2.91 and 3.68 folds compared with the HRGC-HRMS standard method. Conclusion EPM-PCR can be used as an effective means to screen DLCs in fish samples.
