目的应用三维超声探讨兔乳腺VX2移植瘤放射治疗前后的血流变化,研究其与MVD、VEGF表达之间的关系。方法 20只兔建立40个乳腺VX2移植瘤模型,第2周对20个右侧兔乳腺VX2移植瘤进行放射治疗,应用三维超声监测肿瘤生长情况,于治疗前(第2周)、第3周及第4周利用VOCAL-Ⅱ功能量化肿瘤内部及周边的血供,并测量VI值、FI值及VFI值;4周后处死瘤兔,用免疫组化法检测左右侧肿瘤MVD及VEGF的表达,评价三维超声相关检测指标的价值。结果治疗前兔两侧乳腺VX2移植瘤VI值、FI值及VFI值基本一致,第3周及第4周右侧(治疗侧)较左侧(未治疗侧)VI、FI及VFI值减小(P<0.01),且第4周的三维血流参数与MVD及VEGF表达呈正相关。结论三维超声血流参数与MVD及VEGF的表达呈正相关,有助于评价乳腺肿瘤放射治疗的疗效。
Objective To investigate the changes of blood flow before and after radiotherapy in rabbit breast VX2 xenografts using three-dimensional ultrasound, and to investigate the relationship between MVD and VEGF expression. Methods Twenty rabbits were used to establish 40 breast VX2 xenograft tumor models. In the second week, 20 right rabbit VX2 xenografts were treated with radiotherapy. Three-dimensional ultrasound was used to monitor tumor growth before treatment (week 2) and week 3 In the fourth week, the VOCAL-II function was used to quantify the blood supply within and around the tumor, and the VI, FI and VFI values were measured. After 4 weeks, the rabbits were sacrificed and the expression of MVD and VEGF in left and right tumors was detected by immunohistochemistry. , evaluate the value of three-dimensional ultrasound related indicators. Results The VI value, FI value and VFI value of the VX2 xenografts on both sides of the rabbit before treatment were basically the same. The values of VI, FI and VFI were decreased in the right side (the treatment side) and the left side (the untreated side) at the 3rd week and the 4th week. (P<0.01), and there was a positive correlation between 3D blood flow parameters and MVD and VEGF expression at the 4th week. Conclusion The three-dimensional ultrasound parameters are positively correlated with the expression of MVD and VEGF, which is helpful to evaluate the efficacy of radiation therapy for breast tumors.
