目的探讨辛伐他汀对1型糖尿病大鼠心肌组织中核因子NF-κB、肿瘤坏死因子-α(TNF-α)、基质金属蛋白酶-9(MMP-9)表达的影响及其对糖尿病心肌病的作用。方法Wistar大鼠54只,随机分为正常对照组、糖尿病组、辛伐他汀干预组。于实验的4、8、12周分别取6只大鼠,免疫组化和RT-PCR法测定心肌中NF-κB、TNF-α、MMP-9的表达;光镜下观察心肌形态结构变化。结果与对照组相比,糖尿病组和干预组心肌中NF-κB、TNF-α、MMP-9显著增高(p<0.01);而干预组显著低于糖尿病组(p<0.01或p<0.05)。结论糖尿病心肌组织中NF-κB、TNF-α、MMP-9的升高与糖尿病心肌病的发生密切相关,辛伐他汀可以降低心肌中NF-κB、TNF-α、MMP-9的表达,从而对糖尿病心肌病起到保护作用。
Objective To investigate the effect of simvastatin on the expression of nuclear factor-kappa B, tumor necrosis factor-α (TNF-α) and matrix metalloproteinase-9 (MMP-9) in myocardium of type 1 diabetic rats and its effect on diabetic cardiomyopathy effect. Methods 54 Wistar rats were randomly divided into normal control group, diabetic group and simvastatin intervention group. At the 4th, 8th and 12th week of the experiment, 6 rats were taken respectively. The expression of NF-κB, TNF-α and MMP-9 in myocardium were detected by immunohistochemistry and RT-PCR. Results Compared with the control group, the levels of NF-κB, TNF-α and MMP-9 in the diabetic group and the intervention group were significantly increased (p <0.01), while those in the intervention group were significantly lower than those in the diabetic group (p <0.01 or p <0.05) . Conclusion The increase of NF-κB, TNF-α and MMP-9 in diabetic myocardium is closely related to the occurrence of diabetic cardiomyopathy. Simvastatin can reduce the expression of NF-κB, TNF-α and MMP-9 in myocardium Diabetes cardiomyopathy play a protective effect.
