目的研究与铁代谢相关的铁调素(Hepcidin)、骨形成蛋白(BMP6)、铁调素调节蛋白(HJV)在肠癌组织中的表达及其临床意义。方法选取2012—2013年于苏州大学附属太仓第一人民医院进行手术切除的肠癌患者(存档蜡块)62例,另选15例肠癌患者的癌旁正常肠黏膜组织作为对照。应用免疫组化技术对肠癌组织中Hepcidin、BMP6、HJV的表达进行检测和分析。结果肠癌组织中Hepcidin阳性表达率高于正常肠黏膜组织,T_3、T_4期Hepcidin阳性表达率高于T_1、T_2期,差异有统计学意义(P<0.05)。有无淋巴结转移和有无远处转移Hepcidin阳性表达率比较,差异无统计学意义(P>0.05)。BMP6阳性表达率在正常肠黏膜组织中与肠癌组织比较,差异无统计学意义(P>0.05)。T_1、T_2期BMP6阳性表达率高于T_3、T_4期,差异有统计学意义(P<0.01)。有无淋巴结转移和有无远处转移BMP6阳性表达率比较,差异无统计学意义(P>0.05)。HJV阳性表达率在正常肠黏膜组织中与肠癌组织比较,差异无统计学意义(P>0.05)。T分期、有无淋巴结转移和有无远处转移HJV阳性表达率比较,差异无统计学意义(P>0.05)。贫血与Hepcidin、BMP6和HJV表达均无相关性(r=0.08、-0.15、0.07,P>0.05)。Hepcidin与BMP6、HJV表达均无相关性(r=-0.13、-0.07,P>0.05)。结论肠癌组织中Hepcidin、BMP6的表达与患者的T分期有关。肠癌患者术前贫血与肿瘤组织中Hepcidin、BMP6、HJV的表达无关。Hepcidin与BMP6、HJV均无相关性。
Objective To investigate the expression and clinical significance of Hepcidin, BMP6 and HJV in colorectal cancer. Methods Sixty-two patients with bowel cancer who undergone surgical resection (archive wax block) from 2012 to 2013 in Taicang First People’s Hospital affiliated to Soochow University were enrolled in this study. Paraffin-embedded normal paraffin tissues of 15 patients with colorectal cancer were selected as control. The expression of Hepcidin, BMP6 and HJV in colorectal cancer tissues was detected by immunohistochemistry. Results The positive rate of Hepcidin in colorectal cancer tissues was higher than that in normal intestinal mucosa. The positive rate of Hepcidin in T_3 and T_4 was significantly higher than that in T_1 and T_2 (P <0.05). There was no significant difference in the positive rate of Hepcidin between lymph node metastasis and distant metastasis (P> 0.05). The positive rate of BMP6 expression in normal intestinal mucosa compared with that of colorectal cancer, the difference was not statistically significant (P> 0.05). The positive expression rates of BMP6 in T_1 and T_2 were higher than those in T_3 and T_4, the difference was statistically significant (P <0.01). There was no significant difference in the positive rate of BMP6 between lymph node metastasis and distant metastasis (P> 0.05). HJV positive expression rate in normal intestinal mucosa compared with intestinal cancer, the difference was not statistically significant (P> 0.05). T stage, with or without lymph node metastasis and distant metastasis HJV positive expression rate comparison, the difference was not statistically significant (P> 0.05). There was no correlation between anemia and Hepcidin, BMP6 and HJV expression (r = 0.08, -0.15, 0.07, P> 0.05). Hepcidin had no correlation with BMP6 and HJV (r = -0.13, -0.07, P> 0.05). Conclusion The expression of Hepcidin and BMP6 in colorectal carcinoma is related to the T stage of patients. Preoperative anemia in patients with colorectal cancer was not related to the expression of Hepcidin, BMP6 and HJV in tumor tissues. Hepcidin and BMP6, HJV no correlation.
