目的观察鸡脊髓及背根神经节(DRG)中钙离子通道α2(CCα2)亚基的免疫学表达,并观察在DRG中CCα2与calbindin-D28k(CB)的共表达的相互关系。方法通过免疫组织化学,观察了鸡背根神经节中表达情况,利用图像分析系统对VDCC-α2δ-1亚单位免疫阳性和阴性神经元数量和细胞面积进行测量,统计出不同大小细胞的构成比。结果在脊髓背角I、II层和背内侧束(LT)内,CCα2δ亚基免疫阳性呈点状样分布于神经终末之中。在DRG神经元中,约72%的神经细胞呈CCα2免疫阳性,其中小型至中型神经元占免疫阳性神经元的88%。约75%的CCα2免疫阳性神经元与CB共存,其中大部分(90%)神经元是小、中型神经元。结论CCα2主要表达于伤害性神经元中,并在DRG伤害性感觉神经元中,伤害性信息的传递可能与细胞的Ca2+调节机制,即通过CCα2δ亚基的Ca2+内流和细胞内CB的Ca2+结合活性这两种因素有关。
Objective To observe the immunological expression of calcium channel α2 (CCα2) subunit in chicken spinal cord and dorsal root ganglion (DRG) and to observe the relationship between CCα2 and calbindin-D28k (CB) co-expression in DRG. Methods Immunohistochemistry was used to observe the expression of DRG neurons in the dorsal root ganglion. The number of immunoreactive and negative neurons in the VDCC-α2δ-1 subunit of the VDCC-α2δ-1 subunit was measured by image analysis system. The constituent ratios . Results In the spinal dorsal horn I, II layer and the dorsomedial bundle (LT), CCα2δ subunit immunopositive positive dot-like distribution in the nerve terminal. In DRG neurons, about 72% of the neurons were CCα2 immunopositive, with small to medium neurons accounting for 88% of immunopositive neurons. Approximately 75% of CCα2-positive neurons coexist with CB, with the majority (90%) of the neurons being small and medium-sized neurons. Conclusions CCα2 is mainly expressed in nociceptive neurons. In nociceptive DRG neurons, the transmission of nociceptive information may be related to the regulation of Ca2 + in cells, that is, Ca2 + influx through CCα2δ subunit and Ca2 + in intracellular CB Activity of these two factors.
