目的探讨β-arrestin1在儿童急性淋巴细胞白血病(ALL)中的表达及临床意义。方法收集155例初发ALL患儿的骨髓样本(ALL组),分离单个核细胞,以51例非恶性血液疾病患者作为对照(Ctrl组),将ALL患者分成高危、中危和标危组,采用实时荧光定量RT-PCR法检测β-arrestin1 mRNA的表达,采用蛋白质印迹法和免疫荧光法检测β-arrestin1蛋白表达的变化。用Spearman统计分析β-arrestin1表达与ALL患者临床和生物学特点之间的相关性。结果 ALL组β-arrestin1 mRNA和蛋白表达均高于Ctrl组(P值均<0.01)。高危组和中危组β-arrestin1 mRNA表达高于标危组,且高危组β-arrestin1 mRNA表达高于中危组,差异具有统计学意义(P<0.05)。Spearman相关分析显示,β-arrestin1表达与外周血白细胞数(r=0.458,P=0.002)和危险分级(r=0.344,P=0.022)正相关。结论儿童ALL患者骨髓β-arrestin1表达水平与患者的危险分级密切相关,对ALL诊断、治疗策略和预后具有重要意义。
Objective To investigate the expression of β-arrestin1 in childhood acute lymphoblastic leukemia (ALL) and its clinical significance. METHODS: A total of 155 ALL patients with ALL were enrolled in this study. Mononuclear cells (PBMCs) were isolated from 51 patients with non-hematologic malignancies (Ctrl group). All patients were divided into high risk, intermediate risk and standard risk groups, The expression of β-arrestin1 mRNA was detected by real-time fluorescent quantitative RT-PCR. The expression of β-arrestin1 protein was detected by Western blotting and immunofluorescence. Spearman statistical analysis of β-arrestin1 expression and ALL patients with clinical and biological characteristics of the correlation. Results The expression of β-arrestin1 mRNA and protein in ALL group were significantly higher than those in Ctrl group (P <0.01). Β-arrestin1 mRNA expression in high-risk group and intermediate-risk group was higher than that in standard risk group, and β-arrestin1 mRNA expression in high-risk group was higher than that in intermediate risk group (P <0.05). Spearman correlation analysis showed that β-arrestin1 expression was positively correlated with leukocyte count (r = 0.458, P = 0.002) and risk classification (r = 0.344, P = 0.022) Conclusion The expression of β-arrestin1 in children with ALL is closely related to the risk grade of ALL patients. It is of great significance for the diagnosis, treatment strategy and prognosis of ALL patients.
