目的:观察芪附理中灌肠方对溃疡性结肠炎(UC)大鼠血清白介素-8(IL-8)、白介素-10(IL-10)及肠黏膜细胞间黏附分子-1(ICAM-1)表达的影响,以探讨其治疗UC的可能机制。方法:SPF级雄性大鼠36只随机分为模型组、空白组、柳氮磺吡啶组(0.32g/kg)及芪附理中灌肠方组(5.51,11.02,22.04g/kg)共6组。使用TNBS/无水乙醇建造UC大鼠模型,于造模后第8天开始给药,持续灌肠给药14天,观察芪附理中灌肠方灌肠对UC大鼠IL-8、IL-10及ICAM-1表达的影响。结果:与空白组相比。模型组大鼠肠黏膜损伤评分显著增高,较模型组而言,芪附理中灌肠方(22.04g/kg)量组肠黏膜炎症评分显著降低;与空白组相比,模型组大鼠血清IL-8、及肠黏膜ICAM-1含量显著增高,IL-10含量明显显著降低;与模型组相比,芪附理中灌肠方(22.04g/kg)量组血清IL-8及肠黏膜ICAM-1含量显著降低,IL-10含量明显增高。结论:芪附理中灌肠方对UC大鼠肠黏膜具有较好的抗炎修复作用,其机制可能与上调IL-10、下调IL-8及ICAM-1的表达有关。
Objective: To observe the effects of Qifuxie Zhongke Decoction on serum levels of interleukin-8 (IL-8), interleukin-10 (IL-10) and intestinal mucosal intercellular adhesion molecule-1 (ICAM- 1) in rats with ulcerative colitis (UC) ) To explore the possible mechanism of its treatment of UC. Methods: Thirty-six male Sprague-Dawley rats were randomly divided into 6 groups: model group, blank group, sulfasalazine group (0.32g / kg) and Qilifuzheng enema group (5.51,11.02,22.04g / kg) . The UC rat model was established by using TNBS / ethanol. After 8 days of model establishment, UC was administrated continuously for 14 days. Compared with the model group, the score of intestinal mucosa inflammation of the model group was significantly lower than that of the model group (P <0.05), and the intestinal inflammation score of the model group was significantly lower than that of the model group -8, and ICAM-1 in intestinal mucosa were significantly increased and IL-10 content was significantly decreased. Compared with the model group, serum IL-8 and ICAM- 1 content was significantly reduced, IL-10 content was significantly increased. Conclusion: Qifanshichu enema has a good anti-inflammation repair effect on intestinal mucosa in UC rats, which may be related to the up-regulation of IL-10, down-regulation of the expression of IL-8 and ICAM-1.
