本文综述了HBV DNA复制、转录及其调控的研究进展。HBV DNA的复制包括:cccDNA形成,前基因组RNA的合成与包装,DNA负链合成及DNA正链合成等四个主要步骤.HBV DNA转录的机理目前不十分清楚,其可能有赖于宿主体内的RNA聚合酶Ⅱ,转录的模板是cccDNA。目前已经证实HBV DNA可转录3.5kb,2.1kb和0.8kb三种主要的转录体。HBV DNA转录受顺式和反式激活因子调控,HBV增强子成分和启动子成分是主要的顺式激活因子;HBxAg可能是HBV DNA转录的反式激活因子,这些调控因子共同参与HBV DNA转录的调节。
This review summarizes the progress of HBV DNA replication, transcription and regulation. HBV DNA replication includes four main steps: cccDNA formation, synthesis and packaging of pre-genomic RNA, DNA negative strand synthesis and DNA positive strand synthesis.The mechanism of HBV DNA transcription is not well understood at present, which may depend on the RNA in the host Polymerase II, the transcribed template is cccDNA. It has been confirmed that HBV DNA can transcribe three major transcripts of 3.5kb, 2.1kb and 0.8kb. HBV DNA transcription is regulated by cis- and transactivators, and HBV enhancer and promoter components are the major cis-acting activators. HBxAg may be a transactivator of HBV DNA transcription and these regulators are involved in HBV DNA transcription adjust.
