产黄青霉粗提液系由我科实验室制备,经用考马氏亮蓝G250法测定蛋白含量为44~48mg/ml 此粗提液经聚丙烯酰胺凝胶电泳出现3条蛋白带.即P_1、P_2a、P_2b,与标准蛋白比较,分子量约为94 000,43 000及25 000。Sephadex G 200凝胶层析结果呈现两个峰带.经连续收集样品.并按电泳结果分装为3管,即T_1 T_2,T_3,以3管样品对青霉菌致哮喘患者作皮内试验,证实T_2具有最强的生物活性.由于P_2是产黄青霉中T_2的主要部分.从而提出P_2a是产黄青霉中变应原性最强的蛋白成分。
The crude extract of P. chrysogenum was prepared by our laboratory, and the protein content was determined to be 44-48 mg/ml by Coomassie Brilliant Blue G250. This crude extract showed three protein bands by polyacrylamide gel electrophoresis. That is, P_1, P_2a, and P_2b have molecular weights of approximately 94 000, 43 000, and 25 000 compared with standard proteins. The gel chromatographic results of Sephadex G 200 showed two peak bands. Samples were continuously collected and divided into three tubes according to the result of electrophoresis, namely T_1 T_2, T_3. Intradermal test was performed on asthmatic patients induced by Penicillium using three tubes. It was confirmed that T 2 has the strongest biological activity. Because P 2 is the main part of T 2 in P. chrysogenum, P 2a is proposed to be the most allergenic protein component of P. chrysogenum.
