目的:检测特发性血小板减少性紫癜(ITP)患儿血液中B细胞激活因子(BAFF)的表达,探讨儿童ITP的发病机制。方法:采用双抗体夹心酶联免疫吸附试验(ELISA)法检测40例ITP患儿治疗前后及40名对照者血清中BAFF的表达,采用实时荧光定量PCR法检测外周血单个核细胞BAFF-mRNA表达,并进行比较。结果:ITP患儿治疗前血清中BAFF的表达和外周血单个核细胞中BAFF-mRNA表达,均明显高于治疗后和对照者(均P<0.01)。外周血单个核细胞中BAFF-mRNA表达水平与血清中BAFF表达水平呈正相关(P<0.05)。结论:ITP患儿BAFF基因的高表达可能参与了ITP的发病机制。
Objective: To detect the expression of B-cell activating factor (BAFF) in children with idiopathic thrombocytopenic purpura (ITP) and to explore the pathogenesis of ITP in children. Methods: The serum levels of BAFF in 40 ITP children before and after treatment and in 40 controls were detected by ELISA. The expression of BAFF-mRNA in peripheral blood mononuclear cells was detected by real-time fluorescence quantitative PCR , And compare. Results: The expression of BAFF and the expression of BAFF-mRNA in peripheral blood mononuclear cells of ITP children before treatment were significantly higher than those of the control and the control (all P <0.01). The level of BAFF-mRNA in peripheral blood mononuclear cells was positively correlated with the level of BAFF in serum (P <0.05). Conclusion: The high expression of BAFF gene in children with ITP may be involved in the pathogenesis of ITP.
