采用溶胶-凝胶法制备一种蜂胶小分子纳米水凝胶,评价其对口腔溃疡家兔的治疗效果,观察其对口腔溃疡家兔血清白细胞介素-6(IL-6)和肿瘤坏死因子α(TNF-α)表达水平的影响,探明其作用机制。采用40%冰醋酸灼烧法制备家兔口腔溃疡模型,随机分5组,空白组不给予治疗,蜂胶小分子纳米水凝胶低剂量组(GEL低剂量组)、蜂胶小分子纳米水凝胶中剂量组(GEL中剂量组)、蜂胶小分子纳米水凝胶高剂量组(GEL高剂量组)按1支/2次/d麻醉后给药,西瓜霜组按2mg/2次/d麻醉后给药。在治疗过程中测量家兔口腔溃疡直径,观察空白组、GEL高剂量组、西瓜霜组在治疗前和治疗4 d、8 d后口腔黏膜病理组织变化,检测血清IL-6和TNF-α表达水平。结果表明,与空白组比较,GEL低剂量组(P<0.01)、GEL中剂量组(P<0.001)、GEL高剂量组(P<0.001)和西瓜霜组(P<0.001)家兔口腔溃疡直径明显缩小。GEL高剂量组平均愈合时间为(9.5±1.29)d,与西瓜霜组比较,P>0.05,无明显差异性,溃疡部位组织病变明显减轻,治疗效果与西瓜霜组相当。免疫功能失衡是口腔溃疡最重要的发病机制,而IL-6和TNF-α与口腔溃疡免疫失衡有密切关系,GEL高剂量组与西瓜霜组都能使IL-6及TNF-α显著下降,因此其机制可能是通过降低血中IL-6及TNF-α等炎性细胞因子表达水平来促进溃疡的愈合。
A propolis small molecule nano-hydrogel was prepared by sol-gel method to evaluate its therapeutic effect on rabbits with oral ulcer, and its effect on serum interleukin-6 (IL-6) and tumor necrosis factor α (TNF-α) expression levels to explore its mechanism of action. The model of oral ulcer in rabbits was prepared by 40% acetic acid burning method. The rabbits were randomly divided into 5 groups, the blank group was not treated, the low dose of propolis small molecule nano-hydrogel (GEL low dose group), propolis small molecule nano-hydrogel The middle dose group (GEL middle dose group) and propolis small molecule nano hydrogel high dose group (GEL high dose group) were given anesthesia after 1/2 times / d, watermelon cream group was treated by 2mg / 2 times / d After administration. The diameter of oral ulcer in rabbits was measured during the course of treatment. The pathological changes of oral mucosa were observed before treatment, 4 days and 8 days after treatment in the blank group, GEL high-dose group and watermelon cream group, and the levels of IL-6 and TNF- Level. The results showed that compared with the blank group, the low dose GEL group (P <0.01), GEL middle dose group (P <0.001), GEL high dose group (P <0.001) and watermelon cream group Diameter significantly reduced. The mean healing time of GEL high-dose group was (9.5 ± 1.29) d, which was significantly higher than that of watermelon cream group (P> 0.05). The lesions of ulcer lesion were significantly relieved. Immune imbalance is the most important pathogenesis of oral ulcer, while IL-6 and TNF-α are closely related to immune imbalance in oral ulcer. Both GEL high-dose group and watermelon frost group can significantly decrease IL-6 and TNF-α, Therefore, its mechanism may be through the reduction of blood levels of IL-6 and TNF-α and other inflammatory cytokines to promote ulcer healing.
