目的:观察真武汤对心力衰竭大鼠血清半乳糖凝集素-3(Galectin-3)及热休克蛋白70(HSP70)水平影响,探讨其治疗心力衰竭作用机制。方法:通过结扎大鼠冠状动脉前降支配合力竭式游泳成功复制心力衰竭大鼠模型,将造模成功的50只大鼠随机分成中药组(真武汤低、中、高剂量组)、西药组(卡托普利组)、模型组和空白组,每组10只,每日2次灌胃,持续15 d。用酶联免疫吸附试验法(ELISA)法测定大鼠血清BNP、Galectin-3及HSP70水平,并通过电镜观察心肌细胞超微结构。结果:真武汤组与西药组均可明显降低心力衰竭大鼠BNP水平,使Galectin-3及HSP70表达下调,真武汤低、中、高剂量组均优于或等同于西药组,电镜观察显示真武汤组和西药组心肌超微结构均有不同程度改善,真武汤大剂量组最为显著。结论:真武汤调控Galectin-3及HSP70的水平可能是其治疗心力衰竭的作用机制之一。
Objective: To observe the effect of Zhenwu Decoction on the levels of serum galectin-3 and HSP70 in rats with heart failure, and to explore its mechanism of action in treating heart failure. Methods: Fifty rats were randomly divided into Chinese medicine group (Zhenwu Tang low, middle and high dose group), western medicine Group (captopril group), model group and blank group, with 10 rats in each group. Gastrulation was given twice a day for 15 days. Serum levels of BNP, Galectin-3 and HSP70 were measured by enzyme linked immunosorbent assay (ELISA), and the ultrastructure of myocardial cells was observed by electron microscope. Results: Both Zhenwu decoction group and western medicine group could significantly decrease the level of BNP in heart failure rats, and decrease the expression of Galectin-3 and HSP70. Zhenwu decoction group was better than or the same as the Western medicine group by electron microscopy. The ultrastructure of myocardium in both Soup group and Western medicine group improved to some extent, and the Zhenwu decoction group was the most significant. Conclusion: Zhenwu decoction can regulate Galectin-3 and HSP70 levels in the treatment of heart failure.
