目的 观察不同年龄组 (4月和 2 4月龄 )大鼠和连续灌胃给予中药复方 962 3个月的老年大鼠的外周血淋巴细胞DNA的损伤情况。方法 利用快速、灵敏的单细胞电泳技术结合激光扫描共聚焦显微镜来观察大鼠外周血淋巴细胞的DNA损伤 ,以彗星样细胞出现率 (计数一定量细胞中彗星样细胞所占的比例 )、总彗星长度 (“彗星”电泳方向上的最大长度 )和尾距 (尾部DNA占总DNA的百分比与头尾部中心间距的乘积 )来评价淋巴细胞DNA的损伤程度。结果 发现老年大鼠外周血淋巴细胞中彗尾样细胞出现率显著高于青年大鼠 (P <0 0 1) ,复方 962不仅能明显降低老年大鼠的淋巴细胞的彗尾样细胞出现率 ,而且能明显降低老年大鼠外周血彗星样细胞的总彗星长度和尾距 (P <0 0 1)。结论 实验表明复方 962对老年大鼠淋巴细胞DNA的损伤有明显的保护作用。
Objective To observe the DNA damage of peripheral blood lymphocytes in rats of different age groups (April and 24 months old) and those who were given gavage continuously for 962 months. Methods Rapid and sensitive single-cell electrophoresis combined with laser scanning confocal microscopy was used to observe DNA damage in peripheral blood lymphocytes of rats, with the appearance of comet-like cells (the ratio of comet-like cells in a certain number of cells), total The length of the comet (maximum length in the direction of the comet’s electrophoresis) and the tail distance (the ratio of the percentage of DNA in the tail to the total DNA and the distance between the center of the head and the tail) were used to evaluate the extent of DNA damage in lymphocytes. The results showed that the frequency of iris-like cells in peripheral blood lymphocytes of aged rats was significantly higher than that of young rats (P < 0.01). Compound 962 not only significantly reduced the frequency of lymphocyte iris-like cells in aged rats. In addition, the total comet length and tail distance of peripheral blood cometary cells in aged rats were significantly decreased (P < 0.01). Conclusions Experiments show that compound 962 has obvious protective effects on DNA damage in aged rats.
