目的:建立HPLC法同时测定森登-4有效部位中7种成分的含量。方法:采用Extend-C18(250mm×4.6 mm,5μm)色谱柱,以乙腈(A)-0.6%磷酸水溶液(B)为流动相,梯度洗脱,流速0.5 m L·min-1,柱温27℃,检测波长为254 nm和292 nm,进样量10μL。结果:没食子酸、京尼平-1-β-D-龙胆二糖苷、栀子苷、(2R,3R)-双氢杨梅素、芦丁、杨梅素、槲皮素的质量浓度与其峰面积具有良好的线性关系(r=0.999 9);平均回收率为97.30%~103.9%,RSD为0.08%~1.00%。没食子酸、京尼平-1-β-D-龙胆二糖苷、栀子苷、(2R,3R)-双氢杨梅素、芦丁、杨梅素、槲皮素平均含量分别为0.379%、0.447%、2.740%、3.965%、0.253%、0.878%、0.044%。结论:经方法学验证,本法可用于森登-4有效部位的化学成分含量测定。
Objective: To establish an HPLC method for simultaneous determination of seven components in Senden-4 active fractions. METHODS: The mobile phase was eluted with a mobile phase of Extend-C18 (250 mm × 4.6 mm, 5 μm) using acetonitrile (A) -0.6% phosphoric acid (B) as the mobile phase at a flow rate of 0.5 mL · min- ℃, the detection wavelength is 254 nm and 292 nm, the injection volume is 10 μL. Results: The concentrations of gallic acid, genipin-1-β-D-gentiobioside, geniposide, (2R, 3R) -dihydromyricetin, rutin, myricetin and quercetin were (R = 0.999 9). The average recoveries were 97.30% ~ 103.9% with RSD 0.08% ~ 1.00%. The average contents of gallic acid, genipin-β-D-gentiobioside, geniposide, (2R, 3R) -dihydromyricetin, rutin, myricetin and quercetin were 0.379% and 0.447 %, 2.740%, 3.965%, 0.253%, 0.878%, 0.044%. Conclusion: This method can be used for the determination of the chemical constituents of Senden-4 active site by methodological verification.
