目的:探讨重组人淋巴毒素(rh L T) 体内外抗肿瘤作用。方法:体外采用 M T T 法测rh L T 对人胃癌细胞 M K N45 的细胞毒作用,体内采用小鼠移植性肿瘤研究其抗肿瘤活性。结果:rh L T 对体外培养的 M K N45 有较强的细胞毒作用,其 I C50 为1.98×10- 2 μg/ ml;rh L T 对小鼠体内移植的 S180 实体瘤和小鼠 Ehrlich 氏实体瘤均有一定的抑制作用, 其抑制率分别为34 .13 % 和34 .08 % ; 在rh L T 对小鼠黑色素瘤 B16 肺转移的抑制作用研究中发现rh L T 有很好的疗效, 各组的抑瘤率分别为: rh L T 预防组, 30 .61 % ;rh L T 治疗组, 59 .45 % ; rh L T 预防治疗组79.23% 。结论:rh L T 在体内外均有一定的抗肿瘤作用,并有潜在的预防转移作用。
Objective: To investigate the anti-tumor effects of recombinant human lymphotoxin (rh L T) in vitro and in vivo. METHODS: The cytotoxic effect of rh L T on human gastric cancer cells M K N 45 was measured in vitro by the M T T method. The anti-tumor activity of the mouse transplanted tumor was studied in vivo. Results: rh L T had a strong cytotoxic effect on M K N45 cultured in vitro with an I C50 of 1.98×10 −2 μg/ml; rh L T transplanted S180 solid tumors in mice and Mouse Ehrlich’s solid tumors all had certain inhibitory effects, and the inhibition rates were 34. 13% and 34. In the study of the inhibitory effect of rh L T on lung metastasis of mouse melanoma B16, it was found that rh L T had a good effect, and the inhibitory rates of each group were: rh L T prevention group, 30 . 61%; rh L T treatment group, 59 . 45 % ; rh L T prophylaxis treatment group 79.23%. Conclusion: rh L T has a certain anti-tumor effect in vivo and in vitro, and has the potential to prevent metastasis.
