本文应用酶联免疫吸附技术对IL-6/2融合蛋白(FPIL-6/2)进行了定性、定量分析,测定其等电点,并对该蛋白的表位、柔性和抗原性进行了计算机模拟。ELISA测试结果表明:融合蛋白IL-6/2含有IL-2和IL-6两个结构域,可分别与IL-2和IL-6单克隆抗体结合,且两个结合无明显空间排阻效应,这与计算机的表位、柔性和抗原性分析结果一致。从计算机模拟也可以看出FPIL-6/2的抗原性与IL-2和IL-6的一致性,而且接头区抗原性较低,这为该蛋白的实际临床应用提供了一定的保证。
In this paper, the IL-6/2 fusion protein (FPIL-6/2) was detected by enzyme-linked immunosorbent assay. The computer simulation also shows that the antigenicity of FPIL-6/2 is consistent with that of IL-2 and IL-6, and the antigenicity of the linker region is low, which provides a guarantee for the practical clinical application of FPIL-6/2.
