目的 观察表皮生长因子 (EGF)对类风湿关节炎 (RA)滑膜细胞的作用和细胞中有丝分裂素激活的蛋白激酶 (MAPK)的激活情况。方法 3~ 5代体外培养的RA滑膜细胞 ,3H TdR掺入检测EGF对细胞DNA合成的影响 ;EGF刺激后裂解细胞 ,收获蛋白 ,Westernblot检测MAPK的活化。结果 EGF刺激组和对照组每分钟计数值差异有显著性 (P <0 0 0 1)。EGF能引起细胞内明显MAPK活化。结论 MAPK可能是EGF刺激RA滑膜细胞增生的信号传导通路中的重要介质。
Objective To observe the effect of epidermal growth factor (EGF) on rheumatoid arthritis (RA) synovial cells and the activation of mitogen - activated protein kinase (MAPK) in cells. METHODS: The synoviocytes from RA rats cultured in vitro for 3 to 5 days were stained with 3H-TdR to detect the effect of EGF on DNA synthesis. After stimulation with EGF, the cells were harvested, and the protein was harvested. The activation of MAPK was detected by Western blot. Results There was a significant difference in the counts per minute between the EGF-stimulated group and the control group (P <0.01). EGF can cause significant intracellular MAPK activation. Conclusion MAPK may be an important mediator in the signal transduction pathway of EGF-stimulated synoviocyte proliferation in RA.
