以自行合成的胞浆特异性Ca2+荧光探针STDIn-AM标记胃底平滑肌细胞,采用激光共聚焦线扫描技术,首次研究了5-羟色胺诱导胃底平滑肌细胞内钙释放的最基本单位钙火花现象,探讨了其作用机制.表明5-HT与存在于胃底平滑肌质膜上的5-HT2受体结合而使之激活,激活后的5-HT2受体通过Gq/ll蛋白介导的IP3/Ca2+和DG/PKC双信使途径调节钙池内钙释放,内钙释放导致钙池耗竭随即又会促发胞外Ca2+内流;同时,激活的5-HT2受体又直接引起细胞膜去极化,从而激动L型钙通道促发外钙内流.这两种因素促发的胞外Ca2+内流,随即激活质膜附近SR上的ryanodine受体开启单个或多个RyR受体Ca2+通道,从而引起局部内钙释放,形成钙火花.
