用间接免疫荧光方法,将神经肽Y Y1 受体免疫反应物质(LI)定位于大鼠腰段背根节小神经元和脊髓Ⅱ层神经元近细胞膜处。用共聚焦显微术显示背根神经元中Y1 受体-LI呈点状分布于神经元表面。在电子显微镜下发现Y1 受体-LI主要定位于背根节小神经元细胞膜下被膜囊泡、内在小体和管状池,有时也定位于细胞膜上。在脊髓Ⅱ层中,Y1 受体-LI定位于神经元细胞膜,许多树突为Y1 受体阳性,与未被标记的轴突终末形成突触;Y1 受体-LI主要定位于突触后膜以外的细胞膜,只有在少数情况下定位在突触后膜。表明在大鼠背根节小神经元中Y1 受体为胞体受体,并参与受体内在化过程;在脊髓背角神经元中,Y1 受体是连接后/突触后受体。
The indirect immunofluorescence method was used to locate the neuropeptide Y Y1 receptor immunoreactive substance (LI) in the rat lumbar dorsal root ganglion neurons and spinal cord layer II neurons near the plasma membrane. Confocal microscopy showed that the Y1 receptor-L1 in the dorsal root neurons showed a dot-like distribution on the surface of neurons. It was found under the electron microscope that the Y1 receptor-L1 mainly located in the dorsal root ganglion neurons sub-membrane capsule vesicles, inner bodies and tubular pool, and sometimes located in the cell membrane. In the spinal cord layer, the Y1 receptor-L1 locates in the neuronal cell membrane. Many dendrites are positive for the Y1 receptor and form synapses with unlabeled axonal terminals. The Y1 receptor-L1 is mainly located in the postsynaptic Membrane membranes other than the membrane, only in a few cases located in the postsynaptic membrane. It is suggested that Y1 receptor is a cell body receptor in rat dorsal root ganglion neurons and participates in the process of receptor internalization. In spinal dorsal horn neurons, Y1 receptor is a post-junction / postsynaptic receptor.
