目的:NSA2是1种新发现的与细胞增殖相关的核蛋白,本实验旨在初步探讨NSA2是否参与了脉络膜新生血管(Chor-oidal neovascularization,CNV)病变过程。方法:采用532 nm波长的眼底激光对16只小鼠光凝处理,诱导CNV动物模型;小鼠眼底激光光凝处理5 d后,去除眼前段,定量PCR检测NSA2 mRNA;眼球冰冻切片,用NSA2抗体对NSA2蛋白进行间接免疫荧光染色。结果:相对于未经过任何处理的正常对照组,CNV组NSA2 mRNA表达明显上调(P<0.05)。较正常脉络膜组织,CNV组织细胞NSA2阳性率明显增高(P<0.05);CNV区域附近的节细胞NSA2蛋白检测呈阳性,与之相对的正常对照组和CNV组中的远离CNV区域,均未发现NSA2阳性表达的节细胞。结论:NSA2在CNV动物模型眼组织中表达上调,我们推测NSA2可能与CNV病变过程相关联。
Aims: NSA2 is a newly discovered nuclear protein related to cell proliferation. This experiment aimed to investigate whether NSA2 is involved in the pathogenesis of choroidal neovascularization (CNV). Methods: The CNV2 animal model was induced by photocoagulation of 16 mice with 532 nm wavelength fundus laser photocoagulation. After laser photocoagulation in mice for 5 days, the anterior segment of the eye was removed and NSA2 mRNA was detected by quantitative PCR. Antibodies perform indirect immunofluorescence staining of NSA2 protein. Results: Compared with the normal control group without any treatment, the expression of NSA2 mRNA in CNV group was significantly increased (P <0.05). Compared with normal choroidal tissue, the positive rate of NSA2 in CNV tissue cells was significantly increased (P <0.05); NSA2 protein positive in ganglion cells in the vicinity of CNV was detected, while in the normal control group and CNV group, NSA2-positive cells. CONCLUSIONS: NSA2 is up-regulated in CNV animal models of eye tissue. We speculate that NSA2 may be associated with CNV pathological changes.
