本文将简要介绍国内电子显微镜(简称电镜)超薄切片的常规技术,供初学者便于了解和使用这种技术。表1介绍了人的眼睛借助仪器可以观察的范围。在电镜日常工作中,如何提高电镜的分辨率使之达到预期的目的,标本制备工作是十分重要的环节。也是电镜工作中最繁重、最艰巨的工作。这方面国内外均有专著出版。现结合前人的经验和国内一些实验室常用的方法和操作规程加以介绍。
This article will briefly introduce the domestic electronic microscope (referred to as electron microscopy) thin section of the conventional technology for beginners to understand and use of this technology. Table 1 shows the range of human eyes that can be observed with the aid of the instrument. In the daily work of the electron microscope, how to improve the resolution of the electron microscope to achieve the expected purpose, specimen preparation is a very important part. It is also the heaviest and most arduous task in electron microscope work. In this regard, monographs are published at home and abroad. Now combined with the experience of previous generations and some domestic laboratories commonly used methods and procedures to be introduced.
