应用体外血小板聚集实验,观察了3种蝎毒肽(Scirpionvenompeptides,SVP)对凝血酶诱导的家兔血小板聚集抑制作用,以期从蝎毒中筛选具有纤溶和溶栓活性的有效成分。应用0.125,0.25,0.50mg·ml-1各3个剂量组的SVP,SVP2,SVP3对凝血酶0.03U·ml-1(终浓度)诱导的血小板聚集有显著的抑制作用。其抑制率分别为1.51%,1.72%,10.87%;12.01%,25.22%,28.12%;0.94%,6.52%,15.52%。结果提示:蝎毒肽的不同提取物可不同程度地抑制兔血小板聚集,其中以SVP2作用最强,且有量效相关性。
In vitro platelet aggregation assay was used to observe the inhibitory effect of three scorpion peptides (SVP) on platelet aggregation induced by thrombin in rabbits, in order to screen the effective components of fibrinolysis and thrombolytic activity from scorpion venom. SVP, SVP2 and SVP3 in three dose groups of 0.125, 0.25 and 0.50 mg·ml-1 inhibited platelet aggregation induced by thrombin 0.03 U·ml-1 (final concentration). . The inhibition rates were 1.51%, 1.72%, 10.87%, 12.01%, 25.22%, 28.12%, 0.94%, 6.52%, and 15.52%, respectively. The results suggest that different extracts of scorpion peptide can inhibit rabbit platelet aggregation to varying degrees, of which SVP2 has the strongest effect and is dose-dependent.
