目的研究不同浓度碘对成纤维细胞(HSF)增殖及凋亡的影响。方法使用碘离子(终浓度为0~10000μg/L)染毒1×104个/ml的成纤维细胞悬液。于4、12、24、48h后测定细胞增殖活性;于24、48h后测定细胞周期及凋亡,计算细胞增殖指数和细胞凋亡率。结果染毒24h,碘离子浓度≤7000μg/L时具有促进成纤维细胞增殖和抑制细胞凋亡的作用;碘离子浓度>7000μg/L时可抑制成纤维细胞的增殖活性和促进成纤维细胞凋亡。染毒48h,碘离子浓度≤3000μg/L具有促进成纤维细胞增殖和抑制细胞凋亡的作用;碘离子浓度≥7000μg/L可抑制成纤维细胞的增殖活性和促进成纤维细胞凋亡。结论碘对成纤维细胞的细胞增殖和凋亡具有双向效应。
Objective To study the effects of different concentrations of iodine on the proliferation and apoptosis of fibroblasts (HSF). Methods 1 × 104 cells / ml fibroblast suspension was exposed to iodide (final concentration of 0 ~ 10000μg / L). Cell proliferation was measured at 4, 12, 24, and 48 hours later. Cell cycle and apoptosis were measured at 24 and 48 hours, and cell proliferation index and apoptosis rate were calculated. Results Iodine ion concentration> 7000μg / L could promote fibroblast proliferation and inhibit apoptosis when exposed to iodine ion for 24h. When Iodine ion concentration was more than 7000μg / L, it could inhibit fibroblast proliferation and promote fibroblast apoptosis . Exposure to iodine for 48h, iodide ion concentration ≤3000μg / L could promote fibroblast proliferation and inhibit apoptosis; iodide ion concentration ≥7000μg / L could inhibit fibroblast proliferation and promote fibroblast apoptosis. Conclusion Iodine has a bidirectional effect on cell proliferation and apoptosis of fibroblasts.
