目的探讨抗人胰腺癌单克隆抗体SZ-121的制备并对其进行初步鉴定。方法用人胰腺癌细胞株Patu8988作为免疫原,利用杂交瘤技术制备抗人胰腺癌单克隆抗体,应用流式细胞术和免疫组化鉴定抗体的特异性。结果成功获得一株能够稳定分泌特异性抗体的杂交瘤细胞系SZ-121,该抗体与胰腺癌组织呈阳性反应,与胰腺良性病变组织呈弱阳性反应。结论单抗SZ-121对胰腺癌的早期诊断可能具有一定的潜在价值。
Objective To investigate the preparation of anti-human pancreatic cancer monoclonal antibody SZ-121 and its preliminary identification. Methods Human pancreatic cancer cell line Patu8988 was used as immunogen. The monoclonal antibody against human pancreatic cancer was prepared by hybridoma technique. The specificity of the antibody was identified by flow cytometry and immunohistochemistry. Results A hybridoma cell line SZ-121 that can stably secrete specific antibodies was successfully obtained. The antibody showed positive reaction with pancreatic cancer tissue and a weakly positive reaction with benign pancreatic tissue. Conclusion The monoclonal antibody SZ-121 may have some potential value in the early diagnosis of pancreatic cancer.
