目的:建立用高效液相色谱法测定多花蒿药材中阿格拉宾含量。方法:采用回流提取法,以甲醇为提取溶剂。色谱柱为Agilent NH_2(250 mm×4.6 mm,5μm),以乙腈-水(60:40)为流动相,流速为1.0 ml·min~(-1),检测波长为210 nm,柱温30℃。结果:阿格拉宾与其它杂质能够达到有效分离,在0.5~499.0μg·ml~(-1)浓度范围内线性关系良好(r=0.999 9);平均加样回收率为99.5%,RSD为0.6%(n=9)。结论:该检测方法准确可靠,重复性好,可用于批量多花蒿药材中阿格拉宾含量的测定。
Objective: To establish a HPLC method for the determination of agarabin in Artemisia annua. Methods: reflux extraction method, methanol as the extraction solvent. The column was Agilent NH 2 (250 mm × 4.6 mm, 5 μm) with acetonitrile-water (60:40) as the mobile phase at a flow rate of 1.0 ml · min -1. The detection wavelength was 210 nm and the column temperature was 30 ℃ . Results: Agrabin and other impurities could be effectively separated. The linear relationship was good (r = 0.999 9) in the range of 0.5-499.0 μg · ml -1. The average recovery was 99.5% with a RSD of 0.6 % (n = 9). Conclusion: The method is accurate, reliable and reproducible. It can be used to determine the content of agalapin in Artemisia ordosica.
