为研究哪些基因参与低温胁迫诱导烟草提前开花,本研究根据烟草芯片数据和PCR 扩增获得烟草 NtMYB15 基因的全长846 bp CDS 序列.蛋白序列比对结果显示该基因在进化过程中高度保守.将该基因 322 bp 的片段克隆至RNAi 载体pHellsgate12 中,并通过叶盘法将该基因转入烟草K326 中.对两个独立的转基因株系及非转基因植株进行低温胁迫处理,结果表明,与非转基因植株相比,RNAi 转基因植株的开花时期被延迟.因此,本研究将为培育烟草耐早花品种提供有价值的研究信息.“,”To investigate the genes involved in inducing tobacco to flower early flower under chilling stress,the 846 bp coding sequences of tobacco NtMYB15 gene was obtained according to the chip data and PCR amplification in this study.The result of protein sequence alignment showed that NtMYB15 gene was highly conserved among evolution.A fragment of 322 bp was cloned into the RNAi vector pHellsgate12 and the gene was then transformed into tobacco K326 through leaf-disc transformation method.Chilling stress was imposed to two independent transgenic lines and non-transgenic plants.The result indicated that the flowering time of the transgenic plants was delayed compared with the non-transgenic plants.Therefore,the present study could provide new research information for cultivating tobacco variety which was resistant to early flowering.
