目的 :建立测定人血浆中水飞蓟宾浓度的反相高效液相色谱法。方法 :血样在pH 8 5条件下经乙醚提取后 ,采用 μBondapakC18(30 0mm× 4 6mm)色谱柱 ,甲醇 -乙腈 -磷酸盐缓冲液 (pH 3 0 ) (2 9∶2 0∶5 1)为流动相 ,以萘酚为内标 ,检测波长 2 88nm。结果 :水飞蓟宾在 3 6 95~ 2 36 5ng·mL-1范围内线性相关 (r =0 9998) ,方法回收率为 96 5 % ,日内、日间RSD均小于 2 3% (n =3)。结论 :方法简便快速、灵敏度高 ,适用于该药的临床药理研究
Objective: To establish a reversed phase high performance liquid chromatography for the determination of silybin in human plasma. Methods: The blood samples were extracted with diethyl ether at pH 8 5 and the column was eluted with μBondapak C18 (30 0 mm × 4 6 mm) column. The methanol-acetonitrile-phosphate buffer (pH 30) (2 9:20:51) Mobile phase, with naphthol as internal standard, detection wavelength of 2 88nm. Results: The silybin was linearly correlated in the range of 3695-2365ng · mL-1 (r = 0 9998). The recovery rate was 96.5%. The intra-day and inter-day RSD were less than 23% (n = 3). Conclusion: The method is simple, rapid and sensitive. It is suitable for the study of clinical pharmacology
