目的建立离子对HPLC法测定人血中二甲双胍含量的方法。方法采用ODSC18色谱柱(4.6mm×250mm,5μm);以甲醇磷酸二氢钾溶液(含0.03mol·mL-1庚烷磺酸钠,pH4.7)(40∶60)为流动相,流速:1.0mL·min-1,紫外检测波长232nm,柱温37℃。结果离子对HPLC法测定人血中二甲双胍的线性范围0.25~4μg·mL-1,相关系数0.9993。日内差异、日间差异均小于10%,绝对回收率大于80%。结论本法适用于二甲双胍血药浓度测定。
Objective To establish a method for the determination of metformin in human blood by ion-pair HPLC. Methods The mobile phase consisted of ODSC18 column (4.6 mm × 250 mm, 5 μm), methanol-monopotassium phosphate solution (containing 0.03 mol · mL -1 sodium heptanesulfonate, pH 4.7) (40:60) 1.0mL · min-1, UV detection wavelength of 232nm, column temperature 37 ℃. Results The ion-pair HPLC method was used to determine the linear range of metformin in human blood by 0.25-4μg · mL-1 with a correlation coefficient of 0.9993. Differences within the day, day differences were less than 10%, the absolute recovery rate of more than 80%. Conclusion This method is suitable for the determination of metformin blood concentration.
