目的:初步探讨血管生成素-2(Ang-2)和缺氧诱导因子-1α(HIF-1α)在膀胱移行细胞癌中的表达及其与膀胱移行细胞癌病理分级、临床分期之间的关系。方法:选取经临床病理诊断的膀胱移行细胞癌标本64例,正常膀胱组织19例。应用免疫组织化学S-P法检测Ang-2和HIF-1α在膀胱移行细胞癌中的表达情况,并与临床资料相对照进行统计学分析。结果:膀胱移行细胞癌中Ang-2和HIF-1α的阳性率分别为46.9%(30/64)、42.2%(27/64),而正常膀胱组织均无表达。Ang-2阳性率随其病理分级和临床分期增加而升高,G_1与G_3相比,差异有统计学意义(P<0.05);浅表型和浸润型相比,差异有统计学意义(P<0.01)。HIF-1α阳性率亦随其病理分级和临床分期增加而升高,G_1与G_3相比,差异有统计学意义(P<0.05);浅表型和浸润型相比,差异有统计学意义(P<0.01)。Ang-2与HIF-1α的等级相关系数rs=0.498(P<0.01)。结论:Ang-2和HIF-1α过表达均与膀胱移行细胞癌的恶性程度和浸润进展有关。膀胱移行细胞癌中Ang-2和HIF-1α过表达具有正相关关系;可能两者相互作用,通过影响肿瘤血管形成,共同参与膀胱移行细胞癌的发生、发展。
Objective: To investigate the expression of angiopoietin-2 (Ang-2) and hypoxia-inducible factor-1α (HIF-1α) in transitional cell carcinoma of the urinary bladder and its relationship with the pathological grade and clinical stage of bladder transitional cell carcinoma . Methods: Sixty-four cases of transitional cell carcinoma of the bladder and 19 cases of normal bladder tissue were selected by clinicopathological diagnosis. The expression of Ang-2 and HIF-1α in bladder transitional cell carcinoma was detected by immunohistochemical S-P method and statistically analyzed with clinical data. Results: The positive rates of Ang-2 and HIF-1α in transitional cell carcinoma of the bladder were 46.9% (30/64) and 42.2% (27/64), respectively, but not in normal bladder tissues. The positive rate of Ang-2 increased with the increase of pathological grade and clinical stage, the difference between G_1 and G_3 was statistically significant (P <0.05). There was significant difference between superficial and infiltrating types (P <0.01). The positive rate of HIF-1α also increased with the increase of pathological grade and clinical stage, the difference between G_1 and G_3 was statistically significant (P <0.05). There was significant difference between superficial and infiltrative P <0.01). The rank correlation coefficient between Ang-2 and HIF-1αwas 0.498 (P <0.01). Conclusion: The overexpression of Ang-2 and HIF-1α are related to the malignant degree of bladder transitional cell carcinoma and infiltration progress. There is a positive correlation between Ang-2 and HIF-1α overexpression in transitional cell carcinoma of the bladder. It is possible that the two may interact with each other and affect the occurrence and development of bladder transitional cell carcinoma by influencing tumor angiogenesis.
