该文旨在研究比较霍山石斛、铁皮石斛(霍山)、铁皮石斛(云南)、铜皮石斛和河南石斛等5种石斛对四氯化碳急性肝损伤的保护作用。实验将C57BL/6小鼠随机分为空白对照组、模型对照组、5种石斛高(7.5 g·kg~(-1))和低(1.25 g·kg~(-1))2个剂量组。各组连续给药2周,末次给药2 h后,除对照组腹腔注射橄榄油外,其余各组腹腔注射0.5%CCl4橄榄油溶液。16 h后,采集小鼠血清及肝组织,测定小鼠血清中ALT,AST的活性;并检测肝组织中SOD和MDA的水平;计算肝脏指数并观察肝组织病理学变化,荧光定量PCR技术检测肝组织中IL-1β,IL-6和TNF-α的mRNA的表达水平。结果显示,与模型组相比,各石斛组均能降低小鼠血清指标ALT和AST水平,使肝组织中MDA含量下降,SOD活性升高,减轻CCl4对肝脏的病理损伤。此外,霍山石斛肝组织中IL-1β,IL-6和TNF-α的mRNA表达水平显著下调。结果表明,5种石斛对四氯化碳急性肝损伤具有保护作用。但不同产地石斛和不同种石斛的保肝效果有所不同,其中霍山石斛效果最为显著,其他几种石斛保肝效果依次为铁皮石斛(云南)、铁皮石斛(霍山)、河南石斛和铜皮石斛,这可能与不同基源品种石斛化学成分不同有关。
The purpose of this paper is to study the protective effects of five Dendrobium species, Dendrobium huoshanense, Dendrobium candidum (Huoshan), Dendrobium candidum (Dendrobium officinale), Dendrobium candidum and Dendrobium nobile, on carbon tetrachloride-induced acute liver injury. C57BL / 6 mice were randomly divided into two groups: blank control group, model control group, five kinds of Dendrobium high (7.5 g · kg -1) and low (1.25 g · kg -1) . Each group was administrated continuously for 2 weeks. After the last administration for 2 hours, except for the control group, intraperitoneal injection of olive oil and intraperitoneal injection of 0.5% CCl4 olive oil solution. After 16 h, the serum and liver tissues of mice were collected to measure the activity of ALT and AST in the serum of mice. The levels of SOD and MDA in the liver tissue were measured. The results showed that compared with the model group, each Dendrobium group could reduce the levels of serum ALT and AST in mice, decrease the content of MDA and increase the activity of SOD in the liver tissue, and alleviate the pathological damage of CCl4 to the liver. In addition, mRNA expression levels of IL-1β, IL-6 and TNF-α in the liver of Dendrobium huoshanense were significantly down-regulated. The results showed that five kinds of Dendrobium have a protective effect on CCl 4-induced acute liver injury. However, the dendrobium of different origins and different dendrobium have different protective effects on the liver, of which the dendrobium huoshanense has the most obvious effect, and the detoxification activities of the other dendrobium species are Dendrobium candidum (Yunnan), Dendrobium candidum (Huoshan), Dendrobium nobile and Dendrobium officinale , Which may be related to the different base of different species of Dendrobium chemical composition.
