吲哚-3-甲醇通过抑制Survivin表达诱导人鼻咽癌细胞的凋亡

目的探讨吲哚-3-甲醇在体外实验条件下诱导人鼻咽癌CNE1细胞增殖的影响及其诱导CNE1细胞凋亡的作用与可能机制。方法水溶性四氮唑(WST-1)法检测吲哚-3-甲醇在体外对人鼻咽癌CNE1细胞增殖的抑制作用;DAPI荧光染色法直接观察及通过检测线粒体膜电位反映吲哚-3-甲醇诱导的CNE1细胞凋亡现象;蛋白免疫印迹(Westernblotting)方法检测凋亡相关信号分子存活素(survivin)蛋白表达情况。结果吲哚-3-甲醇在体外可以剂量依赖的方式抑制人鼻咽癌细胞增殖,并可以诱导细胞凋亡,且在凋亡过程中,凋亡抑制蛋白survivin表达降低。结论吲哚-3-甲醇在体外可抑制人鼻咽癌CNE1细胞增殖并诱导其发生凋亡,且凋亡的发生可能与survivin表达受抑有关。 Objective To investigate the effect of indole-3-methanol on proliferation of human nasopharyngeal carcinoma cell line CNE1 and its effect on CNE1 cell apoptosis in vitro. Methods Water-soluble tetrazolium (WST-1) method was used to detect the inhibitory effect of indole-3-methanol on the proliferation of human nasopharyngeal carcinoma CNE1 cells in vitro. DAPI staining was used to observe the mitochondrial membrane potential. - methanol induced apoptosis of CNE1 cells; Western blotting was used to detect the expression of survivin protein. Results Indole-3-methanol can inhibit the proliferation of human nasopharyngeal carcinoma cells in a dose-dependent manner and induce apoptosis in vitro, and the expression of survivin was decreased during apoptosis. Conclusion Indole-3-methanol can inhibit the proliferation and induce the apoptosis of human nasopharyngeal carcinoma CNE1 cells in vitro, and the apoptosis may be related to the suppression of survivin expression.
