目的:探索改良高压锅修复法对免疫组化检测ER(Estrogen Receptor,雌激素受体)、Ki-67(细胞增殖活性抗原)两种核抗原表达水平的影响。方法:选取80例乳腺癌组织,分别进行ER和Ki-67两种核抗原在两组不同的修复方法修复后的免疫组化检测。结果:免疫组化采用改良高压锅修复法与传统修复法相比,ER抗原表达水平80例有13例提高,其中有11例由(+)变为(++),2例由(++)变为(+++),(+)和(++)的样本的结果差异有统计学意义(P<0.05),Ki-67抗原表达水平80例有28例提高,其中20例由(<25%)变为(≥25%~50%),有6例由(≥25%~50%)变为(≥50%~75%),有2例由(≥50%~75%)变为(≥75%~100%),总体差异有统计学意义(P<0.05)。结论:与高压锅传统修复法相比,改良高压锅修复法能提高ER、Ki-67抗原的表达水平,且着色程度更强、节省试剂、方便进行多种pH值修复液同时修复,更加实用。
Objective: To explore the effect of modified pressure cooker repair on the expression of two nuclear antigen (ER), estrogen receptor (ER) and Ki-67 (cell proliferation active antigen) in immunohistochemistry. Methods: Eighty cases of breast cancer tissues were selected for immunohistochemical detection of ER and Ki-67 after two different repair methods. Results: Compared with the traditional repair method, the expression of ER antigen in the immunohistochemical method was improved in 80 cases (13 cases), of which 11 cases changed from (+) to (++) and two cases changed from (++) to (+++), (+) and (+ +) samples showed statistically significant differences (P <0.05). Ki-67 antigen expression was increased in 80 cases (28 cases) (≥25% ~ 50%), 6 cases (≥25% ~ 50%) became (≥50% ~ 75%) and 2 cases (≥50% ~ 75%) became (≥75% ~ 100%), the overall difference was statistically significant (P <0.05). Conclusion: Compared with the traditional autoclave repair method, the improved autoclave repair method can improve the expression of ER and Ki-67 antigen, and more coloring, saving reagents, facilitate the simultaneous repair of multiple pH value repair fluid, and more practical.
