目的:研究T细胞分化蛋白(T-cell differentiation protein,MAL)mRNA在口腔鳞状细胞癌(oral squamous cellcarcinoma,OSCC)及配对正常口腔黏膜组织中的表达及意义。方法:采用RT-PCR方法检测30例OSCC患者肿瘤组织及其配对的正常口腔黏膜中MAL mRNA的表达,所得数据采用SAS6.12软件包进行χ2检验和t检验。结果:RT-PCR检测表明,MAL mRNA在30例OSCC和配对正常黏膜的阳性率分别为36.7%(11/30)和86.7%(26/30)(P<0.001);总表达水平分别为1.20±0.18和2.96±0.36,OSCC中表达水平较正常黏膜中表达下调2.47倍(P<0.001);MALmRNA在OSCC中表达下调与患者的性别、肿瘤的临床分期、有无颈淋巴结转移等临床参数无关,但与病理分化程度显著相关。结论:在OSCC的癌变过程中,MAL mRNA表达下调,表明MAL基因可能是OSCC的一个靶基因;MAL在OSCC诊断中具有潜在的应用价值。
Objective: To study the expression and significance of T-cell differentiation protein (MAL) mRNA in oral squamous cell carcinoma (OSCC) and its matched normal oral mucosa. Methods: The expression of MAL mRNA in 30 cases of OSCC and its normal oral mucosa was detected by RT-PCR. The data were analyzed byχ2 test and t test using SAS6.12 software package. Results: The positive rates of MAL mRNA in 30 OSCC and paired normal mucosa were 36.7% (11/30) and 86.7% (26/30) respectively (P <0.001) by RT-PCR. The total expression of MAL mRNA was 1.20 ± 0.18 and 2.96 ± 0.36 respectively. The expression of MAL mRNA in OSCC was 2.47 folds lower than that in normal mucosa (P <0.001). The down-regulation of MAL mRNA in OSCC was not associated with the gender, tumor stage, presence or absence of cervical lymph node metastasis , But significantly correlated with the degree of pathological differentiation. CONCLUSIONS: MAL mRNA expression is down-regulated in the carcinogenesis of OSCC, indicating that MAL gene may be a target gene of OSCC. MAL has potential value in the diagnosis of OSCC.
