目的:探讨通络保肾复方及其拆方对糖尿病大鼠肾损伤内质网应激凋亡通路的影响。方法:将SD大鼠随机分成正常组、造模组,腹腔注射STZ建造糖尿病大鼠模型,造模成功后随机分成模型组、缬沙坦组、通络保肾复方组、中药通络保肾补虚组、中药通络保肾解毒组。喂养6周后测大鼠血糖(GLU)、糖化血红蛋白(Hb A1c)、血浆白蛋白(ALB)、血肌酐(Scr)及肾组织GRP78、p-JNK、Caspase-12蛋白及基因的表达。结果:6周时与正常组比较,模型组GLU、HbA1c、BUN升高(P<0.01);与模型组比较,缬沙坦组GLU、HbA1c、BUN下降(P<0.01);各中药治疗组HbA1c下降(P<0.01)。Western blot、qRT-PCR结果:与正常组比较,模型组大鼠GRP78、p-JNK表达上调(P<0.01);与模型组比较,各治疗组表达下调(P<0.01,P<0.05)。结论:通络保肾复方及其拆方可抑制内质网应激诱导JNK凋亡通路激活,6周时无Caspase-12信号激活。结合中医学方证理论,以方测证,推测糖尿病肾损伤的中医核心病机为本虚标实。
Objective: To investigate the effect of Tonglu Baoshen Compound and its disassembled formulas on the apoptosis pathway of endoplasmic reticulum stress induced by renal injury in diabetic rats. Methods: The SD rats were randomly divided into normal group, model group and STZ-induced diabetic rat model by intraperitoneal injection. The rats were randomly divided into model group, valsartan group, Tongluanbao Decoction group, Tonic group, traditional Chinese medicine Tongluo detoxification group. After six weeks of feeding, the expressions of GLU, Hb A1c, ALB, Scr and renal tissue GRP78, p-JNK and Caspase-12 protein and gene were measured. Results: Compared with the normal group, the GLU, HbA1c and BUN in the model group were significantly increased at 6 weeks (P <0.01). Compared with the model group, the GLU, HbA1c and BUN in the valsartan group decreased (P <0.01) HbA1c decreased (P <0.01). Western blot and qRT-PCR results showed that compared with the normal group, the expressions of GRP78 and p-JNK in the model group were up-regulated (P <0.01). Compared with the model group, the expression of GRP78 and p-JNK in the model group was down-regulated. CONCLUSION: Tonglu Baoshen Compound and its disassembled recipe can inhibit endoplasmic reticulum stress-induced JNK apoptosis pathway activation, and no Caspase-12 signal activation at 6 weeks. Combination of traditional Chinese medicine theory of evidence to square test, presumed diabetic kidney injury of the core of Chinese medicine based on the virtual standard.
