冷冻含水悬浮生物材料的低温电子显微术是高分辨研究生物大分子结构的重要手段。为获得冷冻含水悬浮生物材料的高质量的相位反差图象,使用带有适当尺寸孔径(3—10μm)和分布的微孔碳支持膜是较为理想的。多数作者采用的微孔碳支持膜制备方法(Fukami,A,and Adachi,K.1965,1972:Murray,J.and Ward,R.1987)或较为复杂,或使用特殊的试剂。我们利用了一种使用常用试剂并较为简单的制备方法亦获得了较为理想的效果。微孔碳膜的孔径大小可以控制并有较好的重复性。主要制备步骤如下:
Cryogenic electron microscopy of frozen aqueous suspended biomaterials is an important method for high-resolution study of biological macromolecules. To obtain high quality phase contrast images of frozen aqueous suspended biological material, it is desirable to use a microporous carbon support membrane with a suitably sized pore size (3-10 μm) and distribution. The microporous carbon support membrane preparation method used by most authors (Fukami, A, and Adachi, K. 1965, 1972: Murray, J. and Ward, R. 1987) is either more complicated or uses special reagents. We also used a more commonly used reagents and relatively simple preparation methods have also been more satisfactory results. Porous carbon membrane pore size can be controlled and have good repeatability. The main preparation steps are as follows:
