目的:探讨不同浓度的溶血卵磷脂(LPC)对人视网膜微血管内皮细胞表达糖基化终产物受体的影响。方法:人视网膜内皮细胞加入不同浓度的LPC:(20μmol/L,40μmol/L,60μmol/L)和糖基化终产物,培养24 h,Real-time(RT)-PCR检测糖基化终末产物受体(RAGE)m RNA表达水平,Western blot检测RAGE蛋白表达水平。结果:Realtime(RT)-PCR及Western blot结果表明,LPC的溶度为60 umol/L作用24 h后,RAGE的m RNA及蛋白表达水平明显高于溶度为20μmol/L、40μmol/L。结论:LPC能促进人视网膜内皮细胞RAGE的表达,并且呈剂量依赖性。
Objective: To investigate the effects of different concentrations of lysophosphatidylcholine (LPC) on the expression of advanced glycation end products receptor in human retinal microvascular endothelial cells. METHODS: Human retinal endothelial cells were exposed to different concentrations of LPC (20μmol / L, 40μmol / L, 60μmol / L) and advanced glycation end products for 24 hours. Real-time RT- Product receptor (RAGE) m RNA expression level, Western blot detection of RAGE protein expression levels. Results: Realtime (RT) -PCR and Western blot results showed that the mRNA and protein levels of RAGE in LAGE were significantly higher than those in 20μmol / L and 40μmol / L after LPC was dissolved at 60 umol / L for 24 h. Conclusion: LPC can promote the expression of RAGE in human retinal endothelial cells in a dose-dependent manner.
