目的:测定青天葵野生球茎和组培球茎中氨基酸含量。方法:采用AccQ-Tag法,以AccQ-FlourTM对青天葵野生球茎和组培球茎中17种氨基酸进行柱前衍生;采用Waters 1525HPLC仪器、AccQ-TagTM柱,以AccQ·Tag浓液-水(1∶10)为流动相A,乙腈-水(60∶40)为流动相B进行梯度洗脱,检测波长248nm,柱温37℃,进样量10μL。结果:17种氨基酸的线性回归方程r值均>0.9997,平均回收率为98.7%,RSD为1.8%(n=6);青天葵野生球茎和组培球茎中均含有17种氨基酸,组培球茎中的总氨基酸含量高于野生球茎,但二者差异无统计学意义(P>0.05)。结论:本法快速、简便,结果准确;从氨基酸含量来考察,组织培养的青天葵具有与野生青天葵相同的品质。
Objective: To determine the content of amino acids in wild bulb and tissue culture bulb of Cyclobalanopsis chinensis. Methods: AccQ-Tag method was used to pre-column derivatization of 17 kinds of amino acids in wild corm and wild plant corm. AccQ-FlourTM was used. : 10) as mobile phase A, gradient elution with acetonitrile-water (60:40) as mobile phase B, the detection wavelength was 248nm, the column temperature was 37 ℃ and the injection volume was 10μL. Results: The r-values of 17 amino acids were all higher than 0.9997, with an average recovery of 98.7% and a RSD of 1.8% (n = 6). Seventeen kinds of amino acids were found in both wild and cultivated bulbous corm, In the total amino acid content is higher than the wild bulb, but the difference was not statistically significant (P> 0.05). Conclusion: The method is rapid, simple and accurate. From the amino acid content, the tissue culture of the blue sunflower has the same quality as wild blue sunflower.
