目的 :研究中药白附子的免疫学调节活性。方法 :用3H介入法测定了中药白附子提取物(Rhizomatyphoniiextract,RTE)对小鼠脾细胞和人淋巴细胞的增殖活性 ;用绵羊红细胞花结形成试验分离纯化淋巴细胞后 ,检测了RTE的效应细胞 ;用51Cr游离实验测定了人杀伤细胞和自然杀伤细胞的杀伤活性 ,用ELISA法和生物法研究了单核细胞的细胞因子产生 ,并用3H介入法分析了单核细胞对肿瘤细胞的吞噬功能 ;用层析法分析了白附子的化学成分 ,并用SDS PAGE法测定了其有效成分的分子量。结果 :RTE对小鼠脾细胞和人淋巴细胞的增殖活性有很强的促进作用 ,并有较好的量效关系 ;RTE的效应细胞是T细胞 ;RTE增强人杀伤细胞对肿瘤细胞的特异性杀伤活性和自然杀伤细胞的非特异性杀伤活性、增强单核细胞的细胞因子 (肿瘤坏死因子和白细胞介素 1)产生 ,并增强单核细胞对肿瘤细胞的吞噬功能。白附子的化学成分为 66道尔顿的糖蛋白。结论 :RTE对人的T细胞和单核细胞有免疫增强作用 ,并通过刺激机体的免疫系统杀伤或吞噬肿瘤细胞和外来抗原 ,这些活性可应用于临床调节免疫功能和治疗肿瘤等疾病。
Objective: To study the immunomodulatory activity of traditional Chinese medicine Aconite. METHODS: The proliferative activity of spleen cells and human lymphocytes in mice was determined by 3H intervention method. The effect cells of RTE were detected after isolation and purification of lymphocytes using the sheep erythrocyte rosette formation test. The killing activity of human killer cells and natural killer cells was determined by 51Cr free assay. Cytokine production by monocytes was studied by ELISA and biological methods. Monocyte phagocytosis of tumor cells was analyzed by 3H intervention. The chemical constituents of Aconite were analyzed by chromatography and the molecular weight of the active ingredient was determined by SDS PAGE. RESULTS: RTE had a strong promoting effect on the proliferation of mouse spleen cells and human lymphocytes, and had a good dose-effect relationship; RTE effector cells were T cells; RTE enhanced the specificity of human killer cells against tumor cells. Killing activity and non-specific killing activity of natural killer cells, enhanced production of monocyte cytokines (tumor necrosis factor and interleukin 1), and enhanced monocyte phagocytosis of tumor cells. The chemical composition of Aconite is 66 dalton glycoprotein. Conclusion : RTE has immunoenhancing effect on human T cells and monocytes, and it can stimulate the body’s immune system to kill or engulf tumor cells and foreign antigens. These activities can be used to clinically regulate immune function and treat tumors and other diseases.
