目的:研究温阳化瘀中药复方对糖尿病大鼠脂肪组织抵抗素mRNA表达的影响,探讨温阳化瘀中药治疗糖尿病胰岛素抵抗的作用机制。方法:采用逆转录多聚酶链式反应(RT-PCR)方法测定糖尿病大鼠腹部脂肪组织和下肢骨骼肌间的脂肪组织抵抗素mRNA表达。结果:肥胖的2型糖尿病胰岛素抵抗模型大鼠具有脂肪源性抵抗素mRNA高表达的特点,而腹腔内脂肪组织的表达水平比周围组织骨骼肌肌间脂肪组织的表达水平更高。(P<0.01)温阳化瘀中药复方治疗可以降低脂肪源性抵抗素的mRNA的表达水平。结论:温阳化瘀中药通过降低脂肪源性抵抗素的mRNA的表达水平治疗胰岛素抵抗。
Objective: To study the effect of compound prescription of Wenyang Huayu on the expression of resistin mRNA in adipose tissue of diabetic rats, and to explore the mechanism of Wenyang Huayu herbs in treating insulin resistance of diabetes. METHODS: RT-PCR was used to determine the expression of resistin mRNA in adipose tissue of abdominal adipose tissue and lower limb skeletal muscle of diabetic rats. RESULTS: Obese type 2 diabetic insulin resistance model rats were characterized by high expression of adipose-derived resistin, and the expression level of intra-abdominal adipose tissue was higher than that of skeletal muscle in the surrounding tissues. (P<0.01) Wenyang Huayu compound Chinese herbal medicine can reduce the expression level of adipose-derived resistin mRNA. Conclusion: Wenyang Huayu traditional Chinese medicine can treat insulin resistance by reducing the mRNA expression of adipose-derived resistin.
