目的:探讨飞行时间质谱在生物大分子纯化鉴定中的应用。方法:利用飞行时间质谱仪检测自孕妇尿液中提取和纯化人绒毛膜促性腺激素(HCG)过程中,不同步骤所获样品的分子量分布。结果:随着提纯过程的进行,粗品HCG不断得到净化,质谱图显示信号峰逐渐减少,纯度逐渐提高。结论:飞行时间质谱是一种新型的测定生物大分子分子量分布的仪器,可用于生物大分子的纯化鉴定,具有高灵敏度、高分辨率、快速、重复性好等特点。
Objective: To explore the application of time of flight mass spectrometry in the purification of biological macromolecules. METHODS: The molecular weight distributions of samples obtained at different steps during the extraction and purification of human chorionic gonadotropin (HCG) from the urine of pregnant women were measured by time-of-flight mass spectrometry. Results: As the purification process progressed, the crude HCG was continuously purified, and the mass spectrum showed that the signal peak decreased gradually and the purity gradually increased. Conclusion: Time-of-flight mass spectrometry is a new instrument for determining the molecular weight distribution of biological macromolecules. It can be used for the purification and identification of biological macromolecules. It has the characteristics of high sensitivity, high resolution, fastness and good repeatability.
