目的:观察溃结灵对溃疡性结肠炎(UC)大鼠模型结肠粘膜超微结构动态变化的影响。方法:UC大鼠模型采用TN-BS法,比较3、7、14天各组肠质量指数变化及透射电镜下观察结肠超微结构改变。结果:溃结灵组及柳氮磺吡啶(SASP)组大鼠一般情况较模型组好,恢复较快。电镜下观察,3天模型组大鼠结肠粘膜上皮表面微绒毛减少,变短,扭曲,线粒体肿胀,内容物丢失,细胞器减少,细胞质液化溶解,腺上皮细胞间连接稍松散,肠腺杯状细胞排列紊乱,细胞内颗粒稀少,呈排空状态;7天超微结构有所改善,肠腺杯状细胞排列较整齐,细胞内颗粒有所增加;14天杯状细胞颗粒较丰富,胞吐较活跃;3、7、14天大鼠肠质量指数均明显高于正常组。3天溃结灵组及SASP组大鼠结肠粘膜上皮表面微绒毛稍减少,腺上皮相互连接紧密,肠腺杯状细胞排列整齐,胞内颗粒丰富,胞吐正常;7天杯状细胞胞吐渐变活跃;14天肠腺几乎全由杯状细胞组成,胞吐十分活跃;3、7、14天大鼠肠质量指数与模型组相比均显著降低。结论:溃结灵对TNBS法UC大鼠模型结肠粘膜超微结构有改善作用。
Objective: To observe the effect of Kuijieling on the dynamic changes of ultrastructure of colon mucosa in rat model of ulcerative colitis (UC). Methods: The UC rats were treated with TN-BS method. The change of intestinal mass index was compared between groups on days 3, 7, and 14 and ultrastructural changes of the colon were observed under transmission electron microscope. Results: The rats in the Kuijieling group and SASP group were generally better than the model group, and recovered quickly. Under electron microscopy, the microvilli on the colon mucosal epithelium decreased, became shorter and twisted, the mitochondria swelled, the contents were lost, the organelles decreased, the cytoplasm liquefy and dissolved, and the junction between glandular epithelial cells was loosened slightly. Disorderly arrangement, intracellular particles are sparse and empty; 7-day ultrastructure is improved, goblet cells are arranged more neatly and intracellular granules are increased; 14-day goblet cells are more abundant and exocytotic Active; Intestinal mass index of rats at 3, 7 and 14 days was significantly higher than that of the normal group. 3 days Kuijieling group and SASP group rats slightly decreased microvilli on the surface of the colonic epithelium, glandular epithelium closely connected, glandular goblet cells arranged neatly, intracellular particles rich, exocytosis was normal; 7 days goblet cell exocytosis Gradually active; 14 days of intestinal glands are almost all composed of goblet cells, exocytosis is very active; 3, 7 and 14 days rat intestinal mass index compared with the model group were significantly reduced. Conclusion: Kuijieling can improve the ultrastructure of colon mucosa in UC rats induced by TNBS.
