目的研究大蒜辣素(A llic in)导致肝癌细胞HepG2凋亡的现象及其影响规律。方法运用CCK-8试剂盒研究A l-lic in对HepG2细胞增殖的影响,运用流式细胞仪检测其对细胞凋亡、DNA代谢的影响,运用免疫印迹(W estern b lot)研究肝癌细胞凋亡基因表达的影响。结果A llic in能抑制HepG2细胞的增殖,细胞倍增时间延长为4 d,同时A llic in也能导致细胞出现大量凋亡,实验结果表明其凋亡率最高为4.13%,死亡率达到70.38%。细胞周期实验结果表明A llic in能使大部分细胞处于DNA合成期的S期及G0/G1期,延缓细胞增殖。W estern b lot实验结果表明A llic in能激活细胞凋亡基因提高药物的治疗效果。结论A llic in能抑制肝癌细胞的增殖可导致细胞大量死亡并引起细胞凋亡,并能引起细胞DNA代谢发生紊乱,细胞大部分被阻滞在DNA合成的G0/G1期,细胞分裂被抑制,W estern b lot实验结果表明细胞凋亡基因大量表达,细胞发生凋亡。
Objective To study the effect of Allicin on the apoptosis of HepG2 cells and its influencing rules. Methods The effect of A l-licin on the proliferation of HepG2 cells was studied using CCK-8 kit. The effect of A l-lic in on the apoptosis and DNA metabolism was examined by flow cytometry. The liver cancer cells withered by Western blot (W estern b lot) were studied. The impact of gene expression in death. Results The proliferation of HepG2 cells was inhibited by A llic in, and the doubling time was prolonged to 4 d. At the same time, A llic in also caused a large number of apoptosis. The experimental results showed that the highest rate of apoptosis was 4.13% and the mortality rate was 70.38%. The results of cell cycle experiments showed that A llic in can make most cells in the S phase and G0/G1 phase of DNA synthesis and delay cell proliferation. W estern b lot experimental results show that A llic in activates apoptotic genes and enhances drug treatment. Conclusion A llic in can inhibit the proliferation of hepatocellular carcinoma cells and lead to the death of a large number of cells and cause apoptosis, and can cause the disorder of DNA metabolism. Most of the cells are arrested in the G0/G1 phase of DNA synthesis, and cell division is inhibited. W estern b lot experimental results show that apoptotic gene expression, apoptotic cells.
