目的探讨Nucleostemin(NS,核干细胞因子)基因在非小细胞肺癌(NSCLC)组织中的表达及临床意义。方法采用免疫组织化学SP法检测53例NSCLC组织和15例癌旁正常组织中NS蛋白的表达,并分析与NSCLC患者临床病理特征的关系。结果 NS蛋白在NSCLC中的表达率为54.7%(29/53)显著高于癌旁正常组织中的表达率0%(0/15)(P0.05)。结论NS基因mRNA及蛋白在NSCLC组织中高表达,对肿瘤细胞的恶性增殖起了重要作用,是一个新的有应用价值的肿瘤分子标志物。
Objective To investigate the expression and clinical significance of Nucleostemin (NS) gene in non-small cell lung cancer (NSCLC). Methods Immunohistochemical SP method was used to detect the expression of NS protein in 53 NSCLC tissues and 15 adjacent normal tissues. The relationship between the expression of NS protein and clinicopathological features was analyzed. Results The positive expression rate of NS protein in NSCLC was 54.7% (29/53), which was significantly higher than that in adjacent normal tissues (0/15) (P <0.05). The positive expression rate of NS protein in adenocarcinoma was 65.2% (15/23), which was significantly higher than that in squamous cell carcinoma (46.7%, 14/30) (P <0.01). The poorly differentiated tissue was 77.8% / 18) was significantly higher than that in high and moderate differentiated tissues (42.9%, P <0.05), which was not associated with TNM stage and lymph node metastasis (P> 0.05). Conclusion The high expression of NS gene mRNA and protein in NSCLC tissues plays an important role in malignant proliferation of tumor cells and is a new molecular marker of tumor.
