目的:建立人血浆中别嘌呤醇及其代谢产物氧别嘌醇的高效液相色谱测定方法。方法:色谱柱为Hypersil BDS C_(18)柱(250mm×4.6mm,5μm),流动相为50mmol·L~(-1)KH_2PO_4(pH 4.63)-甲醇(97:3),流速1ml·min~(-1),柱温30℃,紫外检测波长254nm。以阿昔洛韦为内标,血浆样品用高氯酸沉淀蛋白后分析。结果:别嘌呤醇在0.02~5μg·ml~(-1)范围内线性关系良好(r=0.999 2),提取回收率为76.4%~81.4%,方法回收率为98.7%~112.7%;氧别嘌醇在0.02~8μg·ml~(-1)范围内线性关系良好(r=0.999 6),提取回收率为76.3%~80.2%,方法回收率为97.2%~100.2%。结论:本法操作简便、灵敏、准确。
Objective: To establish a HPLC method for the determination of allopurinol and its metabolite oxypurinol in human plasma. METHODS: The column was Hypersil BDS C 18 column (250 mm × 4.6 mm, 5 μm) with a mobile phase of 50 mmol·L -1 KH 2 PO 4 (pH 4.63) -methanol (97: 3) at a flow rate of 1 ml · min ~ (-1), column temperature 30 ℃, UV detection wavelength 254nm. With acyclovir as an internal standard, plasma samples were analyzed after precipitating the protein with perchloric acid. Results: Allopurinol showed good linearity (r = 0.999 2) in the range of 0.02 ~ 5μg · ml ~ (-1), the recovery was 76.4% ~ 81.4% and the recovery was 98.7% ~ 112.7%. The linear relationship was found in the range of 0.02-8 μg · ml -1 (r = 0.999 6). The recoveries were in the range of 76.3% -80.2%. The recoveries were in the range of 97.2% -100.2%. Conclusion: This method is simple, sensitive and accurate.
