本实验采用抗狂大病毒糖蛋白及核蛋白单抗包被、以抗狂犬病毒核蛋白单抗酶结合物作抗体夹心法(SEIA),检测21份接种了可疑狂犬病街毒的小鼠脑组织悬液,并与小鼠颅内接种法(MIT)比较,结果表明SEIA阳性者17份,滴度范围为1:100~1:1280,这17汾样品经MIT检测亦为阳性,二者符合率达100%,其中2份样品由于病毒量少,要经盲传后,接种小鼠才发病,而用SEIA检测一开始即呈阳性,且随着传代病毒量的增加而SEIA检测滴度也上升.表明SEIA快速、敏感、特异,可用于狂犬病毒病的实验室诊断.“,”The sandwich ELISA(SEIA) was developed to detect doubtable rabies street virus in supernatant of mouse brain specimens. The cocktail of McAbs against rabies glycoprotein and nucleocapsid protein were coated in microplates, and the captured antigen was subsequently indentified by adding peroxidase~conjugated McAbs against rabies nucleocapsid. Meanwhile the mouse inoculating test (MIT) by the intra-cerebral route was made to compare with the SEIA. Of the 21 specimens, 17 were positive in both SEIA and MIT. The correlativity of SEIA and MIT is 100%. It is interesting that there were two specimens which were positive in SEIA but negative in MIT initially,and with the subsequently blind passage in MIT,the inoculated mice died of rabies virus and the dose of them in SEIA were increased. The result suggests that he SEIA could be used in diagnosis of rabies in laboratory as its rapidly,sensitively and specifically.
