目的研究Aurora-A在宫颈癌中的表达情况,以及其表达水平与宫颈癌预后的关系。方法选择2007年1月至2009年1月第四军医大学西京医院妇产科宫颈鳞状细胞癌与宫颈腺癌患者组织标本共74例,对照组选取同期保存的该患者癌旁正常组织(经病理检查证实无肿瘤细胞扩散),通过半定量RT-PCR、蛋白印迹实验在细胞学水平对Aurora-A的表达进行检测;通过免疫组化、实时定量PCR的手段明确Aurora-A在宫颈癌及正常癌旁组织中的表达情况;在此基础上将其表达水平与患者临床病理特征进行统计学分析。结果半定量RT-PCR与Western-blotting实验结果均提示肿瘤细胞系中Aurora-A的表达明显高于正常细胞;实时定量PCR提示51.35%(38/74)的组织标本中Aurora-AmRNA由0.088±0.102升高至0.623±0.410(P<0.001),并且与患者的FIGO分期、肿瘤细胞分化水平、是否存在宫旁浸润、是否存在远处转移等因素呈正相关关系。结论 Aurora-A在宫颈癌组织中存在高表达状态,并且与宫颈癌预后之间存在密切关系。
Objective To study the expression of Aurora-A in cervical cancer and its relationship with the prognosis of cervical cancer. Methods A total of 74 cervical squamous cell carcinoma and cervical adenocarcinoma tissue samples of Xijing Hospital of the Fourth Military Medical University from January 2007 to January 2009 were selected. The control group was selected normal tissue adjacent to the patient Pathological examination confirmed the absence of tumor cell proliferation), Aurora-A expression was detected at the cytology level by semi-quantitative RT-PCR and Western blotting. By means of immunohistochemistry and real-time quantitative PCR, the expression of Aurora-A in cervical cancer and Normal paracancerous tissues; on the basis of this, statistical analysis was made on their expression levels and clinicopathological features. Results The results of semi-quantitative RT-PCR and Western-blotting indicated that the expression of Aurora-A in tumor cell lines was significantly higher than that in normal cells. Real-time quantitative PCR showed that Aurora-AmRNA in tissues of 51.35% (38/74) 0.102 to 0.623 ± 0.410 (P <0.001), and positively correlated with the FIGO staging, tumor cell differentiation, presence of uterine infiltration, and the presence of distant metastasis. Conclusion Aurora-A is highly expressed in cervical cancer and has a close relationship with the prognosis of cervical cancer.
