目的:探讨热化疗对荷瘤鼠及口腔癌患者细胞毒性T淋巴细胞(CTL)及热休克蛋白70(HSP70)表达水平的影响,并初步探讨两者之间的相关性。方法:对Balb/c鼠进行热化疗处理,检测CTL活性及HSP70表达变化情况。20例口腔癌患者接受热化疗,于治疗前后行CTL活性检测,并行免疫组化检测HSP70表达情况。结果:热化疗组CTL活性与正常鼠差异无统计学意义(t=1.78,P=0.66),而与高温治疗组、平阳霉素治疗组和不治疗组比较差异有统计学意义,P=0.00。热化疗组HSP70表达明显增多,且FCM提示差异有统计学意义,P=0.00。临床试验中热化疗后口腔癌患者CTL及HSP70较热化疗前明显升高。结论:热化疗能显著提高荷瘤宿主CTL的活性,以及提高HSP70的表达,HSP70可诱导肿瘤细胞表达CTL。
Objective: To investigate the effect of thermochemotherapy on cytotoxic T lymphocyte (CTL) and heat shock protein 70 (HSP70) expression in tumor-bearing mice and oral cancer patients and to explore the relationship between the two. Methods: Balb / c mice were treated with thermochemotherapy to detect the changes of CTL activity and HSP70 expression. Twenty patients with oral cancer underwent thermochemotherapy. CTL activity was measured before and after treatment. The expression of HSP70 was detected by immunohistochemistry. Results: There was no significant difference in the CTL activity between the thermo-chemotherapy group and the normal group (t = 1.78, P = 0.66), but there was significant difference between the CTL activity and the untreated group . The expression of HSP70 in hyperthermia group was significantly increased, and the difference was statistically significant (P = 0.00). In clinical trials, CTL and HSP70 in patients with oral cancer after thermochemotherapy were significantly higher than those before chemotherapy. Conclusion: Chemotherapy can significantly increase the activity of CTL in tumor-bearing host and increase the expression of HSP70. HSP70 can induce the expression of CTL in tumor cells.
